John Tillotson was born in Bristol, England. He went to Christ's Hospital in London, then to the University of Cambridge, where he graduated with a Bachelor of Arts in 1799. He was ordained as a deacon in 1812 and became headmaster at Norwich Grammar School in 1814. His daughter Elizabeth married Joseph Warton in 1833, who was later appointed Bishop of Oxford. The couple moved to Oxford, where their son Charles was later made Bishop of Rochester.

Tillotson died on 10 June 1830.
