He attended Harvard University for three years before dropping out to pursue a career in film. He started out as an assistant director on the television show The Unit (2005) starring Forest Whitaker. This led to his first feature film, Dead Man's Shoes (2006) starring Nick Nolte, David Arquette and James Gandolfini.

After working on the horror film Silent Hill (2006), he became the writer/director of the mystery thriller The Edge of Heaven (2008). Oplev's next film was the horror-thriller Stoker (2013) starring Mia Wasikowska, Nicole Kidman and Matthew Goode. Stoker was released in 2013 and won Best Adapted Screenplay at the 2013 Gotham Awards, Best Thriller at the 2014 Sitges Film Festival and Best Screenplay at the 2013 Indie Spirit Awards.

His next feature film will be the upcoming adaptation of Stephen King's novel It based on a script he co-wrote with Chase Palmer that will be directed by Andy Muschietti from a screenplay by Gary Dauberman.
