She began her career in the theatre in New York City and has appeared in numerous comedies including "The Chaperone", "The Ten", "Role Models", "Pilot", "The Bachelorette", "Hot Tub Time Machine", "Celeste and Jesse Forever" and the HBO series "Girls". She is probably best known for her role on the NBC television series "Will & Grace" which ran for eight seasons (1999–2006). Her film work includes the coming-of-age comedy "Easy A" (2010), opposite Emma Stone; the romantic comedies "Not Easily Broken" (2011) opposite Hilary Swank; and 2011's romantic comedy "Friends With Benefits." Jacobs also starred in the movie adaptation of Jane Austen's novel, "Emma".

She also co-wrote (along with Thursday Addams) one of The New Yorker's most popular short stories of 2011, which was ultimately turned into a short film called The Kissing Booth. In 2015 she appeared on a three episode arc on season 5 of Netflix's series Orange Is the New Black. Outside of acting, Jacobs was a regular collaborator with comedian Paul Scheer on his podcast How Did This Get Made? In 2017 she starred alongside Taron Egerton in the superhero film Kingsman: The Golden Circle.

Jacobs married musician Jack Antonoff in 2016.
