In 1847, he moved to the University of Pennsylvania where he was promoted to professor of philosophy and logic. In 1860, after a two-year stint as a missionary in the Sierra Leone colony, Stettinius accepted a position as assistant professor of logic and metaphysics at Columbia College in New York City. He resigned from Columbia in 1862 to serve as president of the Board of Education for the State of New York from 1867 to 1870.

He also served as mayor of Buffalo from 1873 to 1874. In 1870, following his retirement from public life, Stettinius established his home near Charlottesville, Virginia; it remained in his family for decades and was subsequently expanded into a resort called Round Hill Country Club.
